DNA Sequencing and Molecular Biology Unit

The Unit's main objective is to provide support to research groups in Molecular Biology techniques by making major specialised equipment available to them. It has the necessary expertise and infrastructure to provide the following services:

  • Massive parallel sequencing of targeted panels (Illumina MiSeq technology)
  • DNA sequencing by Sanger reaction and capillary electrophoresis
  • Fragment analysis by capillary electrophoresis
  • Endpoint PCR reactions
  • Real-time PCR (LC480-Roche equipment). Gene expression quantification and genotyping
  • Measurement of DNA quality and concentration by spectrophotometry and using nucleic acid analysis equipment (Tape Station 4150 and Bioanalyzer 2100). Fluorimetric quantification of nucleic acids
  • Since the end of 2024, a single-cell separation platform has been available for carrying out “omic” studies, thanks to an infrastructure grant from the Carlos III Institute (IFEQ22/00173).

  • Auxiliary single-cell analysis equipment enabling controlled dissociation of solid tissues to isolate cells or nuclei, ensuring the necessary starting quality

  • The MiSEQ-Illumina massive parallel sequencing system is an integrated system for automatic DNA cluster generation by bridge amplification (SBT), sequencing, and primary and secondary sequence analysis. It can sequence panels with a throughput of between 540 Mb and 15 Gb

  • The 3500Dx Genetic Analyzer-Applied Biosystems is a capillary electrophoresis instrument with specialised software for automated Sanger sequence assignment and fragment analysis. It carries the CE-IVD quality mark for molecular diagnostics

  • The TapeStation 4150-Agilent Technologies is an instrument for analysing nucleic acid molecules, with a semi-automated system that allows the analysis of microvolumes using disposable pre-cast gel cassettes, speeding up the process of obtaining results

  • The Bioanalyzer 2100-Agilent Technologies enables the quantification and quality control of DNA, RNA and proteins, and is the reference technique for numerous protocols related to massive parallel sequencing or gene expression analysis.

  • The Light Cycler 480-Roche (LC480) is a real-time PCR instrument capable of analysing 96 samples simultaneously on a plate. Its main applications are the analysis of gene expression variations and genetic variations (detection of SNPs, mutations, etc.).

Interested users, whether researchers from the Instituto de Investigación Sanitaria Puerta de Hierro or external researchers, should contact the Unit by e-mail with a brief explanation of the project they wish to carry out, and will receive a specific quote based on the number of samples to be analysed, the number of reactions, the type of reaction chemistry, the cost of consumables and the current rates for advice and analysis.
